Studies on the hydroxylation of vitamin D in man

Abstract

The authors have studied some of the factors influencing vitamin D hydroxylases in man, using two indirect experimental approaches. In the first study they have considered the effect of a long-term treatment with 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) on the serum levels of 25-hydroxyvitamin D (25-OHD) in postmenopausal osteoporosis, a condition in which high serum levels of 25-OHD and low mean levels of 1,25(OH)2D have been observed. In the second study the effects of the infusion of physiological doses of human parathyroid hormone (PTH) on the serum levels of 1,25(OH)2D and 24,25(OH)2D have been investigated. In the first study a decrease in the circulating levels of 25-OHD was observed during 1,25(OH)2D3 treatment. This could be considered as an indirect evidence of an inhibitory action of 1,25(OH)2D3 on 25-hydroxylase: in this view 1,25(OH)2D3 treatment decreases 25-hydroxylase activity, which is higher than normal in postmenopausal osteoporosis due to the low levels of 1,25(OH)2D.
