The influence of serum-supplemented culture media in a transwell migration assay

Abstract

This study determines the effect of serum-supplemented culture media on BV2 microglia migration in a transwell assay. At 24 hours, a 5% foetal bovine serum (FBS) gradient caused BV2 microglia to migrate towards the lower compartment of the transwell apparatus. Interestingly, FBS-supplemented media in the absence of a gradient also resulted in notable microglia migration. Our work cautions against the use of serum-supplemented culture media in a transwell migration assay when using chemoattractants other than FBS. Serum in the media causes migration, therefore, can confound the interpretation when another chemoattractant is used.
